Изучены закономерности индукции синтеза интерлейкина-8 (IL-8) под действием α-дефензинов в культуре различных клеток человека: линии бронхоэпителиальной карциномы A-549, моноцитов и макрофагов, дифференцированных из моноцитов в присутствии М-CSF, эндотелиальных клеток пупочного канатика (HUVEC), эмбриональных клеток почки человека лини НЕК293, миеломоноцитарной линии ТНР-1. Дефензины в концентрации 10-5-10-4М оказывали цитотоксическое действие на клетки линии А-549 в бессывороточной культуре и индуцировали синтез IL-8. Добавление сыворотки полностью подавляло цитотоксический эффект и индукцию синтеза IL-8. Сходные данные получены для клеток линии HEK293 и эндотелиальных клеток.</p><p>В отличие от клеток A-549, HEK293 и эндотелия, добавление сыворотки в культуры моноцитов и макрофагов не подавляло синтеза IL-8 под действием дефензинов в концентрации 10-5-10-4М, несмотря на полное подавление цитотоксичности. Данные указывают на то, что индукция синтеза IL-8 в культуре моноцитов и макрофагов под действием дефензинов может быть обусловлена существованием специфических клеточных рецепторов для дефензинов и, возможно, может наблюдаться и in vivo.</p></abstract><trans-abstract xml:lang="en"><p>Abstract. We explored the effects of defensins on IL-8 synthesis in various human cells, including bronchoepithelial cell line A-549, monocytes, monocyte-derived macrophages differentiated in the presence of M-CSF, HU-VEC cells, HEK293 and THP-1 cell lines. HNP at 10-5-10-4М induced IL-8 production and cytotoxicity in serum-less A-549 culture. The addition of serum abrogated the cytotoxicity along with the induction of IL-8 synthesis. Similar effects were observed in HEK293 cell line and HUVEC.</p><p>The induction of cytotoxicity along with IL-8 production by HNP at 10-5-10-4М was also observed in the serum-less cultures of human monocytes and macrophages. However, monocytes and macrophages retained HNP-induced production of IL-8 after the addition of serum, while cytotoxic effect of HNP was completely inhibited.
