TNFα, IFNγ и IL-4 индуцируют разные варианты активации эндотелиальных клеток линии EA.hy 926. Наибольшие изменения свойств эндотелиальных клеток линии EA.hy 926 наблюдаются в случае их активации TNFα. Под влиянием этого цитокина происходит достоверное повышение секреции IL-8 и MCP-1, а также миграции и адгезии клеток линии THP-1 к монослою эндотелиальных клеток. Несмотря на это, совместное культивирование преинкубированных с этим цитокином эндотелиальных клеток с клетками линии не вызывает достоверных изменений поверхностного фенотипа последних.</p><p>IFNγ и IL-4 оказывают достоверное ингибирующее действие на адгезию моноцитов к эндотелию. При этом, преинкубация эндотелиальных клеток с IL-4 вызывала как усиление секреции IL-8 и MCP-1, так и усиление миграции клеток THP-1. Преинкубация эндотелиальных клеток с IFNγ, напротив, оказывала ингибирующее действие на оба параметра.</p><p>Совместное культивирование клеток THP-1 с интактными и предварительно инкубированными с IFNγ клетками линии EA.hy 926 приводило к незначительному достоверному повышению уровня экспрессии молекул HLA-DR. В случае предобработки монослоя эндотелиальных клеток IL-4 и последующего совместного культивирования их с клетками THP-1 на последних достоверно повышался уровень экспрессии CD68. Выявленные нами единичные изменения поверхностного фенотипа клеток линии THP-1 не коррелировали с изменениями других изученных параметров: экспрессией адгезионных молекул на эндотелии, хемоаттрактантными свойствами этих клеток и степенью адгезии моноцитов.</p></abstract><trans-abstract xml:lang="en"><p>Abstract. We demonstrated that TNFα, IFNγ and IL-4 induce different patterns of endothelial cells activation. The greatest alterations were observed in TNFα threated endothelial cells. Under the influence of this cytokine there was a statistically significant elevation of IL-8 and MCP-1 production by endothelial cells, of THP-1 adhesion and of migration toward endothelial cells monolayer. At the same time we had not observed any immunophenotype alteration of THP-1 cocultured with endothelial cells treated with TNFα.</p><p>IFNγ и IL-4 induced statistically significant inhibition of THP-1 adhesion. IL-4 treated endothelial cells have elevated levels of IL-8 and MCP-1 production and stimulated THP-1 migrations. IFNγ showed inhibition of IL-8 and MCP-1 spontaneous production and THP-1migration.</p><p>Incubation of THP-1 with untreated and IFNγ treated endothelial cells led to significant elevation of HLA-DR expression. Expression of CD68 was significantly higher in case of THP-1 cocultured with IL-4 treated endothelial cells.
