Известно, что в иммунопатогенезе COVID-19 важную роль играют цитокины, в частности, IL-17 и TNFα, являющиеся важными медиаторами воспаления при COVID-19. Анализ особенностей распределения частот аллелей и генотипов генов IL-17A(-197)*G/A, IL-17F(7488)*T/C и TNFA(-308)*G/A у русских Челябинской области с COVID-19 по отдельности показан нами ранее, целью же настоящего исследования было оценить особенности распределения частот встречаемости и параметры сцепления межгенных гаплотипов, образованных однонуклеотидными полиморфными сайтами указанных выше генов у русских Челябинской области с COVID-19. Мы предполагаем, что анализ гаплотипических частот генов провоспалительных цитокинов IL-17A(-197)*G/A, IL-17F(7488)*T/C и TNFA(-308)*G/A может существенно увеличить значимость данных генов в развитии и течении COVID-19. Так, в ходе работы нами впервые определены частоты встречаемости двухлокусных и трехлокусных гаплотипов, образованных однонуклеотидными полиморфными сайтами генов IL-17A(-197)*G/A, IL-17F(7488)*T/C и TNFA(-308)*G/A у русских Челябинской области с COVID-19. Выявлены гаплотипы, которые можно рассматривать как иммуногенетические факторы устойчивости к COVID-19 (IL-17A(-197)*G~IL-17F(7488)*C, IL-17A(-197)*G~IL-17F(7488)*C~TNFA(-308)*G и IL-17A(-197)*G~IL-17F(7488)*T~TNFA(-308)*A) и гаплотипы, повышающие восприимчивость к заболеванию COVID-19 у русской популяции Челябинской области (IL-17A(-197)*G~TNFA(-308)*G, IL-17A(-197)*G~IL-17F(7488)*T и IL-17A(-197)*G~IL-17F(7488)*T~TNFA(-308)*G). В исследование вошли 2 группы русских Челябинской области: больные COVID-19 (137 человек) и контрольная группа (167 человек). Больные COVID-19 проходили лечение в противоковидных больницах города Челябинска в эпидемический период с декабря 2019 года по апрель 2021 года, имели лабораторно подтвержденный анализ на COVID-19 и не имели сопутствующих заболеваний. Группа контроля была набрана из числа потенциальных доноров стволовой клетки ГБУЗ «Челябинская областная станция переливания крови», не имевших антител к возбудителю SARS-CoV-2, прививок от COVID-19 и не состояли в Федеральном регистре лиц, больных COVID-19. Полиморфные варианты исследуемых нами генов, G-197A в гене IL-17A, T7488C в гене IL-17F и G-308A в гене TNFA, определены с помощью аллель-специфичной полимеразной цепной реакции с последующей электрофоретической детекцией результатов в 3%-ом агарозном геле. Статистическая обработка полученных данных была проведена с помощью оценки критерия X2, критерия отношения шансов и 95% доверительных интервалов. Определение частот встречаемости гаплотипов с учетом ошибки среднего и параметров неравновесного сцепления аллелей в локусах было проведено с помощью программы Arlequin (ver. 3.5). Полученные результаты считали статистически значимыми при p&lt;0,05.</p></abstract><trans-abstract xml:lang="en"><p>Cytokines, in particular IL-17 and TNFa, which are important mediators of inflammation in COVID-19, are known to play an important role in the immunopathogenesis of COVID-19. The analysis of the frequency distribution of alleles and genotypes of the IL-17A(-197)*G/A, IL-17F(7488)*T/C and TNFA(-308)*G/A genes in Russians of the Chelyabinsk region with COVID-19 was shown separately by us earlier, the purpose of this study was to evaluate the distribution features the frequency of occurrence and coupling parameters of intergenic haplotypes formed by single-nucleotide polymorphic sites of the above genes in Russians of the Chelyabinsk region with COVID-19. We suggest that analysis of the haplotypic frequencies of the proinflammatory cytokine genes IL-17A(-197)*G/A, IL-17F(7488)*T/C and TNFA(-308)*G/A may significantly increase the significance of these genes in the development and course of COVID-19. Thus, in the course of our work, haplotypes were identified that can be considered as immunogenetic factors of resistance to COVID-19 (IL-17A(-197)*G~IL-17F(7488)*C, IL-17A(-197)*G~IL-17F(7488)*C~TNFA(-308)*G and IL-17A(-197)*G~IL-17F(7488)*T~TNFA(-308)*A) and haplotypes that increase susceptibility to COVID-19 in the russian population of the Chelyabinsk region (IL-17A(-197)*G~TNFA(-308)*G, IL-17A(-197)*G~IL-17F(7488)*T and IL-17A(-197)*G~IL-17F(7488)*T~TNFΑ(-308)*G). The study included 2 groups of Russians from the Chelyabinsk region: COVID-19 patients (those who were treated in anti-covid clinics in Chelyabinsk during the epidemic period from December 2019 to April 2021 had a laboratory-confirmed test for COVID-19 and had no concomitant diseases) and a control group (individuals from the Chelyabinsk Regional Blood Transfusion Station who do not have antibodies to the SARS-CoV-2 pathogen, vaccinations against COVID-19 and are not included in the Federal Register of Persons Infected with COVID-19). Polymorphic variants of the genes we studied were determined using an allele-specific polymerase chain reaction followed by electrophoretic detection of the results in a 3% agarose gel. Statistical processing of the obtained data was carried out by evaluating the X2 criterion, the odds ratio criterion and 95% confidence intervals. The frequency of occurrence of haplotypes was determined using the Arlequin program (ver. 3.5), taking into account the mean error and the parameters of the nonequilibrium coupling of alleles at loci.
