Работа посвящена исследованию влияния дегидроэпиандростерона сульфата (ДГЭАС) на фенотип и свойства дендритных клеток (ДК) здоровых беременных, генерируемых из моноцитов крови в присутствии GM-CSF и IFNa. Показано, что добавление ДГЭАС (10-6М) с первых суток культивирования достоверно не влияет на субпопуляционный состав ДК. В то же время, добавление ДГЭАС на стадии дозревания приводит к увеличению количества зрелых CD83+ и активированных CD25+ДК, а также их предшественников (CDla+). Изменение фенотипа ДК под влиянием ДГЭАС сопровождается достоверным усилением их аллостимуляторной активности и снижением негативного контроля над количеством СD56+СD16+NK-клеток. Таким образом, ДГЭАС стимулирует генерацию, созревание и аллостимуляторную активность ДК, и одновременно ослабляет их негативную регуляцию в отношении количества СD56+СD16+NK-клеток. ДГЭАС-опосредованное изменение фенотипа и функции ДК обсуждается в качестве одного из возможных механизмов нарушения иммунологической толерантности к антигенам плода у беременных с надпочечниковой гиперандрогенией.</p></abstract><trans-abstract xml:lang="en"><p>Abstract. The study deals with evaluation of dehydroepiandrosterone sulfate (DHEAS) effects upon phenotype and functions of dendritic cells (DCs), generated from blood monocytes of pregnant women by means of GM-CSF and IFN-alpha. It was shown, that initial supplementation with DHEAS (10-6 M, day 1 of culture) did not influence the pattern of DC subsets. Meanwhile, addition of DHEAS at the stage of DC maturation (last day of culture) is accompanied by a significant increase in mature CD83+ cells and activated DCs (CD25+), like as their precursors (CD1a+). Furthermore, the DCs generated in presence of DHEAS were characterized by marked allostimulating activity and decreased ability to downregulate the numbers of CD56+CD16+ NK cells. Hence, DHEAS promotes generation, maturation, and allostimulating activity of DCs, along with decreased negative regulation towards CD56+CD16+NK cell amounts.
