TNFα рассматривается как прототип семейства молекул, с одной стороны, играющих важную роль в регуляции нормальной дифференцировки, роста и метаболизма различных клеток, а с другой – выступающих в роли медиаторов воспалительных процессов при различных заболеваниях человека. Интенсивные междисциплинарные исследования биологической роли и терапевтического применения TNFα привели к пониманию его полифункциональности и установлению его ведущей роли в иммунопатогенезе целого ряда заболеваний, что явилось основанием для разработки «антицитокиновой» терапии моноклональными антителами с высокой специфичностью.Цель – осуществить поиск литературы в области изучения структуры и свойств TNFα, а также его роли в норме, патологии и терапииМатериалы и методы: поиск литературы через научные поисковые системы и базы данных. Рассмотрена роль фактора некроза опухолей-α (TNFα) в организме человека в норме и в условиях развития патологий. Описаны возможные TNF-индуцируемые сигнальные пути, а также существующие ингибиторы на основе моноклональных антител для лечения ревматоидного артрита и других аутоиммунных заболеваний. Несмотря на колоссальные знания, накопленные в области изучения биологических функций TNFα в норме и патологии, интенсивные междисциплинарные исследования этого феномена в организме человека являются актуальными и до сих пор. Несомненные успехи в понимании многозначности TNFα как участника в регуляции различных физиологических и патологических процессов в организме человека послужили причиной внедрения биологических препаратов-антагонистов фактора некроза опухолей на основе моноклональных антител, которые оказались высокоэффективными при ревматоидном артрите и являются действенными безопасными лекарственными средствами.</p></abstract><trans-abstract xml:lang="en"><p>Tumor necrosis factor alpha (generally known as cachectin) was noted as a hallmark since the time of its discovery and still retains its position as one of the most intensely studied molecules in the field of biomedicine. TNFα is considered a prototype molecule of the family which, on the one hand, plays an important role in regulating normal differentiation, growth and metabolism of various cells, and on the other hand, acts as mediator of inflammatory processes in various human diseases. Intensive interdisciplinary studies of TNFα biological role and therapeutic use have led to understanding of its polyfunctional effects and establishment of its leading role in immune pathogenesis of diseases, which was the basis for the development of “anticytokine” therapy with highly specific monoclonal antibodies. We aimed for a search of literature in the field of studying TNFα structure and properties, its role under normal conditions, in different diseases and therapy. The literature studies were performed using scientific searching systems and databases. The results are as follows: the role of tumor necrosis factor-α (TNFα) in human body in normal and pathological state was considered. Possible TNF-inducible signaling pathways, as well as existing inhibitors based on monoclonal antibodies for the treatment of rheumatoid arthritis and other autoimmune diseases are  described.
