Системные блокаторы IL-6, основанные на моноклональных антителах к IL-6 или к специфической субъединице рецептора IL-6, уже применяются в клинике и дополнили арсенал биологических лекарств, среди которых наибольшее распространение получили блокаторы Фактора некроза опухолей (TNF). Ревматические заболевания и их экспериментальную терапию можно моделировать на мышах. В настоящей работе сообщается о повышенном уровне системного IL-6 при развитии артрита, вызванного переносом «патогенных» антител, а также об эффектах нейтрализации IL-6 моноклональными антителами против IL-6 мыши. Наши результаты указывают на патогенную роль двух цитокинов, TNF и IL-6, в экспериментальной модели артрита, индуцированного пассивным переносом антител к коллагену.</p></abstract><trans-abstract xml:lang="en"><p>Interleukin-6 (IL-6) exerts important functions on immune regulation. In case of high expression, IL-6 may promote autoimmune disorders, e.g., arthritis. Systemic IL-6 blockers based on monoclonal antibodies against IL-6, or its specific receptor subunit, are already used in clinical settings, adding to a range of known biological drugs, such as, TNF blockers. Rheumatic disorders and their experimental therapy are reproducible in mice. This study revealed systemically increased levels of IL-6 in developing arthritis caused by transfer of pathogenic antibodies, as well as the effects of IL-6 neutralization by monoclonal antibodies against murine IL-6.
